Students use safe saline mouthwash or hair and Chelex® extraction to obtain a sample of their own DNA. Then they amplify a 440-nucleotide segment of a hypervariable region of the mitochondrial chromosome, which contains numerous single nucleotide polymorphisms (SNPs). After gel electrophoresis confirms amplification, the student samples may be sent for sequencing (the cost of sequencing is not included in the price of the kit). Students then compare their SNPs to ancient hominids and to people from different world populations to discover patterns of DNA variation and to analyze theories of human evolution.

Rated 4 out of 5 by from Fix this kit I do this kit every year with my students and it works wonderfully. However, it is very expensive and gives only enough supplies for a class of 25. In my district no one has class sizes that small. My smallest class is 32 students. Please increase the kit to be enough for larger classes. With the price you are charging it isn't unreasonable to get an entire class out of this kit. Make it for a minimum of 35-38 students. If someone has excess that is great but I have to order extra kits just to meet the needs of my class sizes. Thanks!

Rated 3 out of 5 by from Dolan no longer sequences mtDNA The only problem I have with the kit is that the Dolan Learning Center (DNALC) no longer sequences mtDNA. They mention now working with www.genewiz.com for this service, but I don't see any mention of DNALC on the Genewiz site. Carolina needs to redo the instructions for their kit to include a new system for sequencing and access to the DNALC databases (Romanov, Neandertal, etc.) I used the kit last year when DNALC sequenced the mtDNA and it worked well with the database.
