This kit explores the molecular basis of the inherited ability to taste the bitter chemical phenylthiocarbamide (PTC). Students determine their ability to taste PTC using taste paper. They then use safe saline mouthwash and Chelex® extraction to obtain a sample of their own DNA and amplify a 221-nucleotide region of the PTC taste receptor gene. The 2 alleles differ by a single nucleotide, so restriction digestion of the amplified product followed by gel electrophoresis effectively differentiates the 2 alleles. One allele strongly correlates to the ability to taste PTC.

Rated 5 out of 5 by from Great product to use! I have been using this lab kit for several years now and each year it is super easy to follow and the kids love it. It's great to show how just a SNP can cause such a huge difference in a taste. The set up is easy and the directions are straight forward. I do find that some students have trouble understanding the genetics behind it, but the lab provides excellent material for you to go over with the students to help them see what is going on. The one thing I would love to see changed is to add the enzyme HaeIII automatically to the refill kit. Other than that, this is a great product.

Rated 5 out of 5 by from Near fail-proof I use this kit early in the semester in my 200-level genetics course. Students are engaged because it is an experiment on themselves and they get to learn something new about themselves. The kit works great. I rarely have a student fail to get a readable result. It is a little "below" the level I like for a 200-level undergraduate course but I address this by asking them manipulate the data in ways that are not specifically included in the kit. We talk about incomplete dominance (this locus does show some incomplete dominance). We compare our class genotypic and phenotypic frequencies to the overall population and ask if our class is in Hardy-Weinberg equilibrium. In brief: pros: easy and effective, almost always produces results cons: expensive, at a lower level than I would like (but easy to adjust up a few levels)

Rated 5 out of 5 by from Worked Great! I have seen bad reviews of this product from others. It won't work right if you use a 1.2% agarose gel. It isn't enough to separate the bands. I accidentally missed this fact the first time I did it and the lab resulted in one band for all students. When I did it this year and changed it to 2.0% agarose (which the directions say to use) it worked great! Even better...The genotype that the lab showed for the students matched 95% of my students based on their ability to taste. They learned a lot and enjoyed this lab. Definitely more fun than the ALU insertion as it gives some very interesting information.
