Designed to match traditional AP® Biology Lab 6. Students use specially developed pBLU® plasmid in a colony transformation procedure to observe the phenotypic effect of inserting new genes into living bacteria. Transformants appear as white colonies on the ampicillin medium and as dramatically blue colonies on the ampicillin/X-gal medium.

The Teacher Demonstration Kit includes enough materials for 1 demonstration. The 4-Station Kit includes enough materials for 4 teams of students. The 8-Station Kit includes enough materials for 8 teams of students. Perishables Refill replaces consumed items.

Rated 5 out of 5 by from Great Results and Save Educational Dollars with pBLU Refill We make our own LB broths, agars, and calcium chloride for use with the pBLU refill kit. Our Intro to Biology, General Biology, and Microbiology students learn all about gene transformation by performing the pBLU lab experiment. Results are excellent and students are always excited to see the blue colonies of E. coli on the X-GAL positive plates. With this kit refill and by making media ourselves, we're saving money while providing students with a great gene transformation experiment.

Rated 4 out of 5 by from The kit itself is perfect; this plasmid isn't my favorite. The kit works great--the students love knowing that they genetically transformed something. The transformed colonies appear blue on the growth plate, no UV light needed. Overall, my students were more excited about the pGREEN results than this one, since pBLU gives a solid color instead of fluorescence. We also had higher transformation efficiency rates for pGREEN than for pBLU. A good tip to keep in mind is to be conservative in heat-shocking the bacteria. The instructions say to keep the water bath at 42 deg. I've had great results with 40-41, but killed almost all of the bacteria at 43 (luckily a few survived, so I just incubated them twice as long and the students could still see that it worked). If you're new to this lab, make sure not to schedule your perishables delivery too early--the antibiotic (ampicillin) can break down--and the instructions are clear that you can't prepare the growth plates more than 10 days in advance.

Rated 4 out of 5 by from Worked very well, missing one control group. We got much better results with this transformation than with GFP or Firefly Luciferase genes. I substituted real micropipettes for the plastic ones supplied, but otherwise followed the directions. The instructions and handouts were much better than ones I've gotten from other companies. The experimental everview with minimal instructions (mostlyamounts, times and temps# with line drawings was helpful during the experiment when you don't want to hunt through lots of text in the detailed instructions. Saved me from making a cheat sheet. My only complaint is that the kit has a redundant negative control group and is missing a different control group. The LB Amp -Plasmid and LB Amp XGal -Plasmid groups tell you the same thing #cells without plasmid die on Amp). There is no LB XGal -Plasmid group to show that the bacteria didn't have the the B Galactosidase gene before transformation.

Rated 5 out of 5 by from Exceptional Sucess Rate I strongly recommend that you use the pBlu transformation for AP Bio Lab 6A, as students are immediately able to determine which colonies have expressed the new genes (the blue color is quite visible). Even with less than perfect lab technique, over the last 7 years at least 85% of my lab groups have achieved expression of both new genes for ampicillin resistance and B-galactosidase.
