All materials and instructions for (1) conducting complementation (cis-trans) spot tests to place unknown T4rll mutations in either cistron A or B, and for (2) conducting recombination spot tests to determine by deletion mapping the A-segments within which the rll mutations occur. Exercises are recommended for an individual, a team of 2 or 3 students, or use as a demonstration. Other students, however, can participate when they are given sets of result plates for personal analysis and conclusions. Results are obtained from 4 spot tests and 2 control plates. During the course of the experimental procedure described, up to 8 additional sets of 6 plates each can be easily prepared for distribution to other students. Kit provides enough phage and bacteria for such duplication of results, but the required agar plates and soft agar must be purchased separately. Kit ships with perishable materials. Note: Please refer to Bacteriophage Media and item #124730 Plaque Assay Plating Set. Refrigerate coliphage until use; do not freeze. Keep bacteria culture and agar at room temperature.
