Bringing genetic engineering and protein purification into your classroom has never been simpler. Students get the opportunity to create GFP-expressing bacteria, which glow in the presence of UV light, and then isolate and purify the GFP from the E. coli cells. Intermediate/Advanced - Modules 1 and 2 require some background knowledge. Module 3 is for more experienced classes and requires some technical skill.

These kit modules are great for teaching:

  • The molecular basis of heredity
  • The relationship between genes and proteins
  • How E. coli is used to engineer proteins useful in medicine and manufacturing
  • Three techniques - transformation, protein purification, and protein gel electrophoresis - commonly used in biological research and its areas of application

The lab is set up in 3 modules that can be purchased separately and used in conjunction with one another.

  • Module 1, the Green Gene Colony Transformation Kit (item #211080, #211080P, #211082, or #211082P), is necessary for creating the GFP-expressing bacteria used in the protein purification exercise in Module 2.
  • Module 2 (item #211072 or #211072P) contains all of the reagents and instructions necessary for purifying GFP from E. coli.
  • Module 3 (item #211073 or #211073P) contains the acrylamide gels and other electrophoresis reagents necessary for electrophoretic analysis of the purified GFP. The 3rd module is entirely optional but greatly enhances the teaching value of the lab.

The reagents and instructions are truly student-proof - there will be transformants so long as the students are truly careful. Transformation efficiencies are good and the students enjoy the fluorescent results.

Rated 2 out of 5 by from Items missing When this kit was ordered, it was missing a few things, the LB broth, directions, etc. We did get the plates, the pGreen, and the Ecoli. This was for student use for a project, and it was very frustrating for both of us that all of the components were not included.

I also was unaware that perishables had to be ordered after the kit arrived and all of my organisms that I planned to feed this transformed bacteria for my experiment died.

Rated 5 out of 5 by from Exciting lab for engaging students! This kit has everything you need to do bacterial transformation with a plasmid. I've tried a few plasmids with my AP Biology students, and this one consistently gives very clean results. It's also my students' favorite plasmid! One tip is to be conservative in heat-shocking the bacteria. The instructions say to use a 42 deg. water bath. I've had great results if the water bath was 40-41 but killed most of the bacteria if it was 43 (luckily a few survived, so I just incubated them twice as long and the students could still see that it worked). Make sure you have a UV light source (blacklight), which is not included. The bacteria fluoresce green when successfully transformed, and the students get very excited about their "glowy" bacteria. The transformed colonies will keep fluorescing for several days after transformation, so I was able to bring it to a school showcase a few days later--it made a great impression on prospective students!
