Decarboxylase broth is primarily used to differentiate bacterial groups within the Enterobacteriaceae. Our formulation of the decarboxylase broth starts out a bright purple. If dextrose is fermented by the sample, the medium will shift to yellow, as acid is produced. If the production of acid stimulates the production of decarboxylase in the bacteria, decarboxylation of the amino acid, lysine, results in the formation of amines. This increases the pH and causes a change from yellow to purple. Samples that cannot ferment dextrose, such as Flavobacterium capsulatum (item #155083), will remain purple and won’t turn yellow. Organisms that ferment dextrose, but are not able to decarboxylate lysine, such as Proteus mirabilis (item #155239A), will turn yellow and remain yellow. This indicates that they are lysine negative. If they are positive for lysine, like Escherichia coli (item #155065), they will first turn yellow and then revert to purple.
